ABC技术利用生物素和卵白素具有*亲和性的生物学特征，将卵白素和生物素化辣根酶按照一定的比例混合，形成ABC复合物。该法亦被称作三步法，*步为biotin化二抗和一抗结合，第二步为avidin（亲和素，A液）和二抗上的biotin结合，第三步为HRP偶联的biotin（B液）和avidin结合，而HRP再催化底物显色完成染色。
